Here we show the generation and assembly from human pluripotent stem (hPS) cells of subdomain-specific forebrain spheroids: human cortical spheroids (hCS) and human subpallial spheroids (hSS). This protocol describes hPS cell maintenance, neural spheroid generation, patterning and assembly to obtain functionally-integrated human forebrain 3D cultures.
