_Candida albicans_

This is a protocol to induce disseminated candidiasis in mice. Candida albicans infection experiments were performed as previously described with minor modifications (Blood. 2000 Jun 15; 95(12):3725-3733).

Reagents:

Candida albicans (ATCC 18804).

Sabroud Dextrose Broth (Sigma Aldrich, St Louis, MO).

Sterile phosphate buffered saline (PBS), room temperature.

1 ml syringe.

27-gauge needle.

14 ml round bottom conical tube with snap cap.

Hemocytometer.

Make a frozen stock of subcultures of Candida albicans at -80°C in Sabroud Dextrose Broth containing 15% glycerol.
Plate cultures on Sabouraud dextrose agar plates and grow at 37 °C for 24 hours. The plates can be stored at 4 °C for a maximum of 4 weeks.
Before each experiment, pick a colony from a plate and grow in 5 ml of Sabouraud Dextrose Broth in a 14 ml conical tube at 37 °C for 24 hours in a shaker.
Centrifuge the cells (1000 rpm for 5 minutes) and wash the pellet twice with 5 ml of sterile PBS and resuspend in PBS.
Count the cell number using a hemocytometer, and then adjust to the desired concentration.
Inject 2-5 × 10⁵ colony-forming units of C albicans dissolved in 200 μl of sterile PBS from tail vein to induce disseminated candidiasis.
